The inventors have developed a strategy for purification of post-translationally modified peptides or proteins, and the identification of the various modifications. The methodology involves solid-phase immobilization of cysteine/thiol groups using mixed disulfides of resin-bound alkyl and aryl thiols. The method was utilized for the analysis of numerous cellular cysteine-modification including S-nitrosylation and S-palmitoylation.
